Production and Characterization of Monoclonal Antibodies to Borrelia burgdorferi, the Lyme Disease Agent









Abstract
Borrelia burgdorferi is the causative agent of Lyme disease, but it is difficult to detect B. burgdorferi specifically because of cross-reactivity of B. burgdorferi with Leptospira spp. and Treponema pallidum in the reaction of sera from patients
in
enzyme-linked immunosorbent assay(ELISA) or in indirect immunofluorescent(IFA). In an attempt to develop B. burgdorferi-specific detection, this study was initiated to produce monoclonal antibodies to B. burgdorferi and to use the antibodies in
detecting B. burgdorferi. In this study, twelve monoclonal antibodies specific to B. burgdorferi were produced; ten antibodies(seven were IgG1 and three were IgG2b in isotype) were reactive with 31/34kDa) molecules and two antibodies(IgG1 in
isotype)
were reactive with 41kDa) molecule im immunoblotting. These antibodies were well reactive with B. burgdorferi in IFA, but not reactive with Leptospira interrogans serovar lai, canicola, and T. pallidum I ELISA. Although further evaluations of
these
antibodies are required to know the reactivity to other Borrelia spp., this study suggests that the monoclonal antibodies to 31/34kDa and 41kDa molecules of B. burgdorferi may be used in detecting B. burgdorferi from the clinical specimens and
the
vectors.
